Studying Extrachromosomal Genetic Elements in Sulfolobus

چکیده

Membrane vesicles released by cells are responsible for various cellular functions of Archaea. They can be, for example, a stress response and transport of toxic compound from cells. Although studies have shown that membrane vesicles produced by euryarchaeal Thermococcales could participate in DNA transfer between cells at high temperature (Gaudin et al., 2013), the biochemical compositions and genetic functions of membrane vesicles produced by S. solfataricus P2 remain unclear. Here, we show that the Sulfolobus membrane vesicles contain cellular DNA. Furthermore, DNA sequencing reveals that vesicle-bounded DNA is constituted of random genome segments, but rich in fragments of IS elements. In addition, pyramidal structures were observed in the cells of Solfataricus P1-pKEF9 integrated with SSV2.
